Hepatoprotective Activity of Binahong (Anredera cordifolia (Ten.) Steenis) Leaf Extract in Diabetes Mellitus Rats

Binahong (Anredera cordifolia (Ten.) Steenis) is efficacious in treating diabetes, heart disease, and high blood pressure. This study aims to determine secondary metabolite levels using UV-Vis spectrophotometry and evaluate antioxidant activity which has a positive effect in repairing liver damage caused by streptozotocin. The methods in this research were UV-Vis and DPPH spectrophotometry, the Kruskal-Wallis advanced test, and the Mann-Whitney test on the livers of diabetes mellitus rats. The results of qualitative analysis of binahong leaf extract contain Alkaloids, Flavonoids, Saponins, and Tannins. In the quantitative test, the total flavonoid content was 0.2433% w/w, saponin 0.1631% w/w, tannin 0.2512% w/w, and alkaloid 0.0350% w/w, and the antioxidant activity of the ethanol extract of binahong leaves was obtained The IC50 value of 61.9 ppm is included in the strong antioxidant category. In histopathology, the test animals were induced with streptozotocin and given treatment, except for normal controls, negative control Na-CMC 0.5%, positive control metformin, the test group was given extract at doses of 25, 50, and 100 mg/kg BW. Binahong leaf extract has hepatoprotective activity at a dose of 25 mg/kg BW, which is a level that is effective in repairing damage to rat liver cells with an average damage value of 1.4.
