Description:
Cordia sebestena L. contains flavonoids, that play a role in free radical scavenging activity. This study
aimed to analyze the flavonoids in the hydrolyzed fraction of the 70% ethanol extract of Cordia leaves.
The study was divided into 7 stages: (1) maceration using 70% ethanol, (2) acid hydrolysis of the
ethanol extract, (3) fractionation of hydrolysate with ethyl acetate, (4) determination of total flavonoid
content, (5) antiradical activity assay against 2,2?-diphenyl-1-picrylhydrazyl (DPPH), (6) thin layer
chromatography (TLC) analysis, and (7) identification of flavonoid using UV-Visible Spectrophotometry
and Infrared Spectroscopy. The total flavonoid content of the ethyl acetate fraction was 8.20 mgQE/g
with an IC50 value of 296.91 g/mL against DPPH radicals. There is one spot of flavonoids in the ethyl
acetate fraction (hRf = 85.5). The UV-Vis spectrum on the spot obtained absorption at 340 nm (band
I) and 240 nm (band II). The infrared spectrum shows a signal at 3363 cm-1 (O-H stretching), 1727
cm-1 (C=O), and 1060 cm-1 strong (C-O). The ethyl acetate hydrolysate fraction of Cordia leaf contains
flavonoid similar to the chalcone group. Further analysis is needed using 1
H NMR or FTIR data from
a known chalcone as a standard to confirm the flavonoid
